purpose of usage:

This kit is used to determine the content of zeatin nucleoside (ZR) in plants and related samples .

Experimental principle

The kit uses an indirect method to determine the level of plant zeatin nucleoside (ZR) in the specimen . A microplate was coated with purified plant zeatin nucleoside (ZR) antibody to prepare a solid phase antibody, and a known concentration of plant zeatin nucleoside (ZR) standard was added to the microwell of the coated monoclonal antibody and unknown. Concentration of plant zeatin nucleoside (ZR) sample to be tested, after incubation, biotin-labeled anti- IgG antibody is added, and then combined with streptavidin- HRP to form an immune complex, after thorough washing and adding substrate TMB color development. TMB in HRP enzyme blue, yellow and eventually converted to the action of an acid. The color depth is positively correlated with the plant zeatin nucleoside (ZR) in the sample . The absorbance ( OD value) was measured with a microplate reader at a wavelength of 450 nm, and the concentration of plant zeatin nucleoside (ZR) in the sample was calculated from a standard curve .

Specimen requirements

1 . Samples containing NaN3 could not be detected because NaN3 inhibited horseradish peroxidase ( HRP ) activity.

2. The specimens should be extracted as soon as possible after collection, and the extraction should be carried out according to the relevant literature. The experiment should be carried out as soon as possible after extraction. If you can't test it right away, you can put the specimen in -20 °C Save, but should avoid repeated freezing and thawing

Steps

1.        The number of slats required is determined by the number of samples to be tested plus the number of standards. Multiple holes are recommended for each standard and blank hole. Each sample is determined according to its own quantity, and it is possible to use the double hole as much as possible to make a double hole.

2.        Adding samples: Set blank holes separately (the blank control holes are not added with samples, the other steps are the same), the standard product holes, and the sample holes to be tested. Then add 50 μl of the standard into the well of the standard , add 10 μl of the sample to be tested first, then add 40 μl of the sample dilution (the final dilution of the sample is 5 times) , cover the sealing membrane, and gently shake and mix. 37 °C Incubate for 45 minutes.

3.        Dosing: 20 times concentrated washing solution diluted with distilled water 20 times and used

4.        Washing: Carefully remove the sealing film, discard the liquid, dry it, fill each well with the washing solution, let stand for 30 seconds, then discard it, repeat 4 times, and pat dry.

5.        Biotin-labeled anti- IgG antibody: 50 μl of biotin-labeled anti- IgG antibody was added per well . 37 °C Incubate for 30 minutes

6.        Wash: Operate the same as 4 .

7.        Chain plus streptavidin -HRP: 50 μl added to each well of streptavidin-HRP avidin, mix gently shaken, 37 °C Incubate for 30 minutes.

8.        Wash: Operate the same as 4 .

9.        Color development: Add 50 μl of developer A to each well , then add 50 μl of developer B , gently shake and mix. 37 °C Protected from light for 15 minutes .

10.    Termination: 50 μl of stop solution was added to each well to terminate the reaction (when the blue color turned yellow).

11.    Measurement: The absorbance ( OD value) of each well was measured in sequence with a blank air conditioner of zero and a wavelength of 450 nm . The measurement should be carried out within 15 minutes after the addition of the stop solution .

Precautions

1 . The kit should be taken out from the refrigerated environment and should be used at room temperature for 15-30 minutes before use. If the enzyme label is unsealed after opening, the slats should be stored in a sealed bag.

2 . Concentrated washing liquid may crystallize out. When diluted, it can be heated and dissolved in a water bath. The washing will not affect the result.

3 . The sampler should be used for each step, and the accuracy should be corrected frequently to avoid test errors. It is best to control the loading time within 5 minutes. If the number of specimens is large, it is recommended to use a gun.

4.   If the content of the substance to be tested in the specimen is too high (the OD value of the sample is larger than the OD value of the first hole of the standard well ), first dilute the sample by a certain multiple ( n times) and then measure it. When calculating, multiply by the dilution factor ( × 5 × n ).

5.   The sealing film is intended for single use only to avoid cross-contamination.

6 . The different batch components of this reagent must not be mixed. Reagent B should be stored away from light.

7 . Strictly follow the instructions of the manual, the test results must be based on the microplate reader reading .

8 . All samples, washings and various wastes should be treated as infectious materials.

9. If it is different from the English manual, the English manual shall prevail.
