The activated peroxidase is horseradish peroxidase (HRP), whose natural carbohydrate (sugar) has been slightly oxidized by periodate to produce an amine-reactive aldehyde group. These carbonyl groups of active peroxidase spontaneously and efficiently crosslink with primary amines on antibodies or other proteins. This method is more efficient than other amine reactive chemicals, such as glutaraldehyde coupling, which often results in polymerization and a higher degree of coupling inactivation. Pre-activated enzymes eliminate the inherent difficulties of preparing and verifying activated peroxidases from scratch.

Tips:
* The antibody to be conjugated is preferably dissolved in an ammonia-free buffer (eg MES, MOPS, HEPES, PBS) at a concentration of 10-50 mM and a pH of 6.5-8.5.
* Commonly used non-buffering salts (such as sodium chloride), chelating agents (such as EDTA) and sugar have no effect on coupling efficiency. We do not recommend HRP coupling in the presence of sodium azide or sodium azide in HRP-bound antibodies. Even a glycerol concentration of up to 50% does not interfere with the coupling efficiency.
* The ideal molar ratio of antibody for labeling is: antibody: HRP = 1:4 to 1:1. (Antibodies at concentrations of 0.5-4 mg/ml usually work best.)
HRP Labeling Kit Features:
- Activated HRP-iodate treatment, aldehyde-activated horseradish peroxidase, can be directly coupled to a primary amine site (eg, lysine) on an antibody or other protein.
- Highly active HRP – enzyme activity up to 120-200 units/mg; activated enzymes are stable for at least 12 months at -20 °C.
- Flexible dosage – every 1 milligram of activating enzyme is enough to react with 1 milligram of IgG.
